Description

Tacrolimus is a potent calcineurin?inhibitor immunosuppressant used extensively in research to study T?lymphocyte activation, cytokine regulation, and transplant rejection models. Originally derived from Streptomyces tsukubaensis, tacrolimus binds FKBP?12 to inhibit calcineurin, reducing IL?2 and other cytokines, thus suppressing T-cell mediated immune responses.

These capsules are the PROGRAF®-branded formulation, each containing 1?mg tacrolimus. Manufactured by Astellas Ireland Co. Ltd and packaged in China by Astellas, under NMPA approval number J20150102. Not intended for clinical use—strictly for laboratory research.

Mechanism of Action & Research Applications

Tacrolimus works by forming a complex with FKBP?12, which then inhibits calcineurin phosphatase activity. This action prevents NF?AT and NF??B activation and suppresses cytokine production such as IL?2, IL?4, IL?6, TNF??, and interferon??.

In laboratory research, tacrolimus capsules are used to:

  • Model transplant rejection and prevention

  • Study T-lymphocyte activation and signaling pathways

  • Evaluate immunosuppressive effects in cell or animal models

  • Investigate cytokine regulation and immune modulation

Side Effects (For Reference Only in Experimental Models)

In controlled research settings, tacrolimus may cause:

  • Suppression of T-cell activation

  • Altered cytokine expression

  • Potential nephrotoxicity or neurotoxicity at high exposures

  • Dose-dependent effects on systemic metabolism

These are experimental observations and not indicative of clinical safety.

Disclaimer

Tacrolimus Capsules (PROGRAF®) are for laboratory research use only and not intended for human or veterinary therapeutic use. Use must adhere to institutional biosafety and research safety protocols.
