Mechanism of Action
Lidocaine and prilocaine are amide-type local anesthetics that block voltage-gated sodium channels in nerve membranes, preventing action potential initiation and propagation. The eutectic mixture allows effective skin penetration due to its low melting point.
Research Applications
Experimental dermatology: Used to numb skin before minor procedures (venipuncture, biopsies, laser therapy) in animal or human skin models .
Onset & duration: Analgesia begins ~60 minutes after application and lasts 1–2 hours post-removal; mucosal application acts faster (5–10 min) .
Pharmacokinetics:
Lidocaine: ~70% plasma protein binding, t½ ~1.5–2 h
Prilocaine: 55% protein binding, t½ ~10–150 min
Safety Profile & Handling
Adverse Events (preclinical/human data):
