Mechanism of Action

Lidocaine and prilocaine are amide-type local anesthetics that block voltage-gated sodium channels in nerve membranes, preventing action potential initiation and propagation. The eutectic mixture allows effective skin penetration due to its low melting point.

Research Applications

  • Experimental dermatology: Used to numb skin before minor procedures (venipuncture, biopsies, laser therapy) in animal or human skin models .

  • Onset & duration: Analgesia begins ~60 minutes after application and lasts 1–2 hours post-removal; mucosal application acts faster (5–10 min) .

  • Pharmacokinetics:

    • Lidocaine: ~70% plasma protein binding, t½ ~1.5–2 h

    • Prilocaine: 55% protein binding, t½ ~10–150 min

Safety Profile & Handling

Adverse Events (preclinical/human data):

  • Local: transient blanching, erythema, edema, rash

  • Systemic (rare): dizziness, headache, methemoglobinemia in infants (<0.5%)

Handling Tips:

  • Apply a thick layer under occlusion for 1 hour (>2 hours for deeper analgesia)

  • Avoid broken skin or mucosa to reduce absorption risk

  • Use PPE; dispose as pharmaceutical waste
